Vampires are magically reanimated corpses who are inhabited by the spirits of the deceased and who closely resemble a living person. Vampires feed and survive on the blood of the living, typically on human blood, and they can transform other humans into more of their kind. Vampires are one of the many known supernatural species in The Vampire Diaries and The Originals universe.

A human who has consumed vampire blood and subsequently died with it still in their system will be magically revived as a one of the undead. The nature of the human's death is irrelevant; however, the body must be more-or-less intact and able to function once they reawaken afterward. Furthermore, the person must be mortally and irreversibly wounded or ill, since the vampire blood in their system will only heal whatever non-fatal wounds or one of a majority of illnesses that the victim sustained prior to dying. Following their mortal death, the undead body will awaken in a state of transition. In order to complete the transition, the soon-to-be-vampire must consume a significant amount of human blood, at least a sip's worth, within the next twenty-four hours, or else they will die. Vampires can only feed on human blood to complete their transition, though once they complete this process, they can then choose to feed on blood from other sources, such as the blood of animals, or even that of other vampires. Until a vampire drinks human blood to complete their transition, they will be not be susceptible to the standard weaknesses of their species (i.e. they can still walk in sunlight and can enter homes without an invitation).

As far as the general public of the supernatural community knows, humans can be changed into vampires, and werewolves can be changed into hybrids, though it requires them to either have died with the blood of the Original hybrid, Niklaus Mikaelson, or the blood of his daughter, Hope Mikaelson, in their system in order to accomplish this. Those werewolves who die with Klaus' blood in his system will need to complete their transition into hybrids by feeding upon the blood of a human Petrova doppelgnger (the same blood that was used in the spell to create the Original vampires); if the hybrid tries to complete their transition with any other form of human blood, they will be transformed into an unstable version of a hybrid that will ultimately die an agonizing death. However, werewolves who die with Hope's blood in their system can be successfully turned into hybrids by also feeding on Hope's blood after they awaken to complete their transition, as she is the first of her kind (a true vampire-werewolf-witch hybrid) whose blood possesses different mystical properties than her father's. Witches can also be turned into vampires; however, since Nature has deemed witches as their servants and consider vampires to be abominations, Nature is of the belief that having the powers of both would be such an imbalance of power that it which it would not stand for it. As a result, one cannot be both a witch and a vampire, and those witches who die with vampire blood in their system will lose their connection to magic upon waking in transition. Although, in extremely rare cases, former witches have regained their magic after they were were cured of vampirism, such as in the case of Katherine Pierce after she ingested Qetsiyah's cure for immortality.

Typical vampires are all descended from those who are known as the Original vampires, whom, unlike the majority of vampires, were turned not by dying with vampire blood in their system, but were instead created by a powerful spell fueled by dark magic. However, the ritual that resulted in their transformation was similar to that of normal vampires in that they had to consume enchanted blood (in their case, the blood of the human Petrova doppelgnger Tatia) before being killed with that blood in their system. Afterward, they fed on the blood from another human from their village and were officially transformed into the world's first true vampires, from which all other vampires were created. Their blood possesses mystical properties that are passed down to each and every one of the vampires they sire and their own progeny, connecting all vampires to one of the six Original vampires from whose bloodline they came. Because of this link, the death of an Original vampire will result in this connection being severed, and every vampire in the deceased Original's bloodline will follow them in death within an hour, as demonstrated by the deaths of Finn and Kol Mikaelson, which subsequently caused everyone turned from their bloodline to die as well.
